Hi, I have some questions as I am new to Jellyfin.
I am running Plex on a Windows 11 NUC. Can Plex and Jellyfin coexist without problems on the same machine?
Can Jellyfin play ISOs natively?

They can coexist. A lot of people do this.
Jellyfin has limited support for ISOs. It MIGHT work, but it is best to extract the folder structure or convert to MKV.
